Biochemical studies for the in vitro selection of rice varieties with tolerance to water stress

Abstract
This research paper was carried out in order to make biochemical studies for in vitro selection of water stress tolerance rice varieties with the aim of looking for possible markers which may be useful to genotypic discrimination against such abiotic factor. The percentages of damages measured by cellular membrane permeability were calculated in callus from rice varieties (Perla de Cuba, Jucarito-104, Amistad-82 and INCA LP-7) having different water stress degrees at 24 and 72h of incubating with solutions at different PEG-6000 levels (0, -0.5, -1.0, -1.5 MPa). After 72h of incubating calluses at -1.0 MPa, electrophoretical analyses developed in a discontinued system of polyacrilamide gels of the isoenzymes: perodixase, fosfatases and esterases as well as, peroxidase activity, total protein and proline contents were determined. The result showed differential response of varieties at -1.0 MPa having present its tolerance degree being useful the PEG concentration and some biochemical characters to detect water stress tolerance.
