Cultivation of horse herpes virus type 1 on chicken embryos

Abstract
Introduction. Rhinopneumonia is a widespread disease of horses, both in the world and in Ukraine. It causes significant economic losses to the horse breeding, which leads to loss ofreproductive capacity of mares, to the culling of valuable pedigree animals and to significant costs for the implementation of veterinary and sanitary measures.The goal of the work. To cultivate herpes virus of horses (I-type) on chicken embryos.Materials and methods. The strains SV-69 and M (W) of the rhinopneumonia virus were investigated. To infect 12-day-old embryos viral material was injected into the chorioallantoic cavity at a dose of 0.5-1.0 сm3. The presence of the virus in organs and tissues was determined by cytopathic effect on the monolayer of chick embryo fibroblasts (CEF) and determined the hemagglutination titre (HA) using horse and rabbit red blood cells.Results of research and discussion. The conducted studies revealed that the titer of hemagglutination was high in extracts of the brain, liver and heart of the embryos infected with virus strains, as with the red blood cells of the horse and ram it was within 1: 16 - 1: 64. During the fourth passages of the tested strains on chicken embryos, it was found that the strain M (W) accumulated in the tissues of the embryo but did not cause embryos death in any passage. At the same time, the increase of the SV-69 strain concentration was accompanied by the death of embryos. By the manifestation of the cytopathic effect of the isolated samples on the culture of cells, similar results as in hemagglutination, test have been observed.Conclusions and prospects for further research. Studied some biological properties of reproduction of horses’ rhinopneumonia strains in chicken embryos, namely the dynamics of their accumulation in the organs of infected embryos. It was found that the highest concentration of the virus was in the brain, liver and heart, while in other organs its content is insignificant.
