Design of Reversed Phase Chromatography (RPC) Column Packing Material for Protein Separation: Part 1- Synthesis and Characterization

Abstract
In the first part of the two-part study, the column packed materials in the form of monodisperse and porous particles were synthesized with regard to test their column performances in Reverse Phase Chromatography (RPC). Particles in the form of poly(vinyl acetate-co-divinylbenzene) [poly(VAc-co-DVB)] and poly(divinylbenzene) [poly(DVB)] were produced by “multi-stage micro-suspension polymerization”. The surface of particles were converted to “hydroxyl” form by base hydrolysis that were used as poly(vinylalcohol-co-divinylbenzene) [poly(VA-co-DVB)] particles, having appropriate structural properties like surface and porosity for chromatographic applications. Particle characterization was assessed by Scanning Electron Microscopy (SEM), BET surface area and porosity measurement system and Fourier Transform Infrared Spectroscopy (FTIR).
