Nano Phytosome Quercetin Ameliorates Cognitive Impairment Induced by Maternal Isolation in Rat Pups

Abstract
Introduction: Autism is a kind of neurodevelopmental disorder that the etiology of one is not known, but several genetic and environmental factors increase the risk of spectrum of disorders. Daily maternal isolation during the perinatal life enhance the risk of neuropsychiatric disorders and impairment in cognitive behavior. Quercetin is flavonoid that is widely exist plant-derived foods. Although the quercetin plays an important role as antioxidant, its usage the pharmaceutical field is limited because of their concentrations are very low to cross blood-brain. In this study, to solve this problems, nano phytosome of quercetin is used to maximize output in therapy of ADS. Method: Male and female rats were mated with pregnancy determined. Newborn rats categories in several groups. All groups except control group exposure to single separation from their mother for 3 H per day from postnatal days 1-9. After the lactation period, rats except ADS group were treated by Quercetin (10 and 40 mg/kg) and nano phytosome of Quercetin (10and 40 mg/kg) for 21 days. During the 36-40 days after their birth we perform novel object test. Results: We demonstrated that daily maternal isolation in perinatal life lead to autistic-like symptoms. The discrimination index for novel object test in ASD group was meaningfully lower than control group (P<0/001). nano phytosome quercetin (10 mg/kg) and nano phytosome quercetin (40 mg/kg) treatment for 30 days significantly (P<0/05) was significantly increased in compare to autism group. Discussion: In the present study we used to maternal separation to create autism-like model. We reported that the lack of communication of mother with newborn rats during the perinatal life lead to increase the neuropsychiatric disorders such as memory impairment over its lifespan. Here, the rat pups were isolated from their mothers for 3 h per day from postnatal day 1_9. The result of behavioral study illustrated that maternal-separated rat pups showed low discrimination index in their novel object test. However, nano phytosome quercetin (10 and 40 mg/kg) treatment increased the discrimination index and improved memory impairment.
