Optimization of Process Conditions for Bovine Antimicrobial Peptides Prepared by Microbial Fermentation Method

Abstract

In order to extract antimicrobial peptide from bovine bone and explore the optimum process conditions of fermentation process, bovine bones was taken as raw material, and the bovine bone antimicrobial peptide was prepared by use of mixed fermentation method. The strains selected for mixed fermentation were Lactobacillus acidophilus ( LA) , Streptococcus thermophilus ( ST) and Bacillus cereus ( BC MBL13-U) . Through orthogonal test. The mass concentration of collagen peptide was taken as the index. The optimum nutrient media components are identified as: V ( BC MBL13-U) ∶ V ( LA) ∶ V ( ST) = 1∶ 2∶ 1, sucrose dosage ( mass fraction) is 2. 0%, and bone powder dosage ( mass fraction) is 4. 5%. The central composite design test was conducted on the basis of orthogonal test. The bacteriostatic rate was taken as the index. The optimum fermentation process conditions is identified as: inoculation quantity ( mass fraction) is 4. 0%, fermentation temperature is38 ℃, fermentation time is 42 h, and initial p H is 7. 0. After verification test, under the optimum fermentation process conditions, the highest bacteriostatic rate of polypeptide liquid is ( 95. 03 ± 0. 55) %. The results of bacteriostatic test show that bovine bone antimicrobial peptides have obvious inhibition effect on both Escherichia coli and Staphylococcus aureus.
