Phytochemical analysis and Antimicrobial activities of different plant parts of Vitex negundo L

Abstract
In the present work leaf and bark extracts of V. negundo were screened for antibacterial and anticandidal property. Qualitative phytochemical analysis, agar cup method, micro-dilution method for MIC, TLC bioautography were used as prescreening methods to study antimicrobial properties against eleven Gramnegative, five Gram-positive pathogenic bacteria and four Candida species. One way ANOVA was carried out to find out the existence of significant differences in the total activity (TA) and zone of inhibitions between each solvent extracts. Furthermore, Duncans post hoc test was carried out to determine the difference in the TA and zone of inhibitions. All five extracts (Petroleum ether, chloroform, ethanol, methanol and water) exhibited significant activity with zone of inhibitions 9.6-15.6 mm for bark and 9.3-14.3 for leaf. Ethanol extracts were best for both bark and leaf with MIC (313 to 2500 µg/ml) for leaf and (313 to 5000 µg/ml) for bark. Considering the quantity extracted from bark powder, the total activity was highest for ethanol and lowest for chloroform. TLC bioautography showed zone of inhibition (>20 mm) with R f va lue of 0.72 for e thanol ext r a c t aga ins t S. flexneri. Preliminary phytochemical analysis exhibited presence of carbohydrates, triterpenoids, tannin and phenolic compounds, glycosides, steroid and sterols and flavonoids in different extracts.
