Protocol for the in vitro selection of potato plants resistant to Alternaria solani Sor culture filtrate.
Journal Title: Biotecnología Vegetal - Year 2009, Vol 9, Issue 1
Abstract
The use of culture filtrate of Alternaria solani in the differentiation of susceptible and resistant potato genotypes has been described in scientific literature. However, few researches take into account the role of isolates of A. solani and other factors affecting the production of culture filtrate to use in vitro selection as a tool. This allows plant breeders to reduce the time to obtained new varieties of potato. This study aimed to establish a protocol for in vitro selection of potato plants resistant to early blight with the use of culture filtrate of Alternaria solani Sor, in support to the potato Genetic Improvement Program. The proposed protocol related aspects such as: obtaining of culture filtrate, characteristics of A. solani isolates and plant material, as well as, the descriptive scale for assessing the effect of culture filtrate on potato plants (Solanum tuberosum L.) var. Desirée. The protocol can be used as a useful tool in early stage trials to evaluate the resistance to A. solani in potato genotypes obtained by any method of genetic improvement to this disease. This contributes to increased efficiency of selection in natural conditions. The number of individuals to evaluate in field decreases. In these conditions other factors affect the differentiation of susceptible and resistant genotypes.

Purification of phytotoxic metabolites from the filtrate culture of Fusarium oxysporum f. sp. cubense gcv 01210 (race 1)
Panama disease, caused by Fusarium oxysporum f. sp. cubense, is considered a destructive disease of economic importance in the genus Musa. The culture filtrates of the pathogen have been used to differentiate cultivars...
Control of bacterial contamination in artificial sugarcane seed through early detection methods
The studies for stablishment of artificial seed technology in sugarcane (Saccharum spp hybrido) have not been free of damage produced by bacterial contaminations, principally in the stage before somatic embryos encapsula...
Differential response of three Phaseolus vulgaris L. cultivars to in vitro water stress induced by PEG-6000
The growth and agricultural yield of common beans (Phaseolus vulgaris L.) is affected, among other factors, by limitations in the availability of water. For this reason, there are working on the search for cultivars tole...
Effect of different protocols for applying a magnetic field (0.03T) on growth, viability and pigment composition of Haematococcus pluvialis Flotow in sufficiency and absence of nitrogen
The effect of a magnetic field (0.03T) on growth, cellular viability and pigments accumulation in Haematococcus pluvialis was studied in order to validate potentialities for market. Magnetic field favored growth and cel...
Genetic stability among in vitro eggplant clones induced by different plant growth regulators
Many factors may influence the genetic stability of cloned in vitro plants, among which are the genotype and the regenerative process induced by plant growth regulators. The resulting somaclonal variations may be usefu...
