The evaluation of regeneration ability of two flax ([i]Linum usitatissimum[/i] L.) cultivars in in vitro culture

Abstract
Regeneration possibility of shoots, calli and roots from cotyledon and hypocotyl explants of two flax cultivars, i.e. Selena and Modran, from the Institute of Natural Fibre in Poznan were evaluated. These cultivars differed in the moment of changing from vegetative to reproductive developmental stage (Selena at 16 cm and Modran at 14 cm height). The hypocotyl and cotyledon explants were collected from 6 days old seedlings and incubated on MS (Murashige and Skoog 1962) medium supplemented with 1 mg·l-1 BAP (6-benzylaminopurine) or 1 mg·l-1 NAA (naphthaleneacetic acid) and 1 or 3 mg·l-1 AgNO3 (silver nitrate) separately or BAP and NAA combined with AgNO3. The most crucial influence on the regeneration abilities had genotype, explant type and media components. The cultivar Modran which begins developmental reproductive stage at lower plant height showed higher regeneration abilities in comparison to the cultivar Selena. It was observed that relatively the best medium for regeneration was MS medium supplemented with BAP. The hypocotyls explants showed much better regeneration than cotyledon explants.
