Design and evaluation of diffusion controlled extended release matrix tablets of Ropinirole hydrochloride

Abstract
The present work was aimed to develop an extended release matrix tablet formulation of Ropinirole hydrochloride by Melt Granulation technique using lipid waxes like stearic acid and hydrogenated castor oil in combination using different diluents such as Lactose and MCC. The granules were evaluated for micromeritic properties and tablets were evaluated for weight variation , hardness , friability , drug content uniformity and invitro drug release. The granules showed good flow properties and all tablets demonstrated compliance with compendia requirements. The tablets were subjected to dissolution studies. The drug release study revealed that FT4 formulation containing balanced amounts of stearic acid and hydrogenated castor oil produced a desired release profile of Ropinirole for 24 hours which was considered as an optimized formulation . The effect of diluents was studied on FT4 with same wax concentration which revealed drug release from the tablets decreased with Lactose (FT5) when compared with MCC (FT6). The DSC studies were also indicating the absence of strong interactions between the components and suggesting drug- excipient compatibility in all the formulations examined .All the formulations FT1 to FT6 followed both first order release mechanism and Higuchi's root time kinetics and exhibited anomalous transport mechanism. It may be concluded from the present study that the slow controlled and complete release profile of Ropinirole over a period of 24 hours was obtained from matrix tablets FT4 formulation.
