Abstract

The present study concerns the preparation, characterization and in-vitro antibiotic release studies of various hydroxyapatite (HAp)-antibiotic implantable minipellets for bacterial bone infections like osteomyelitis. Various antibiotics like ciprofloxacin, ofloxacin, gatifloxacin and amoxicillin were loaded into HAp-system by precipitation technique. The significantly high drug loading was investigated in case of ciprofloxacin (64.14 ± 0.99 % w/w) in comparison of others as we followed the same process parameters. Various HAp-antibiotic minipellets were prepared by compressing synthesized HAp-antibiotic powders. Characterization of this drug delivery was done by P-XRD and FT-IR spectroscopy. These minipellets showed sustained release of antibiotics for several weeks and the drug release patterns followed by Higuchi model. This methodology may prove effective to develop high drug loaded various HAp-antibiotic implantable bone-delivery systems, thereby facilitating individualized chemotherapy for local delivery in bacterial bone infections.
