Preliminary study of the effect of PD-L1 on the biological behavior of vascular endothelial cells in keloids

Abstract

Objective To clarify the difference in the expression of PD-L1 in vascular endothelial cells of keloids and normal skin tissues, and to clarify the regulatory effect of PD-L1 on vascular biological behavior through in vitro function experiment. Methods Clinical samples of keloid were collected, its vascular structure was observed by HE staining, the expression of PD-L1 was detected by Western blotting and PCR, and the expression level of PD-L1 in tissues was observed by immunohistochemistry and immunofluorescence staining. Immunofluorescence was used to determine the position relationship between inflammatory cells and vascular endothelial cells in keloid tissues. The primary extracted T cells were co-cultured with PD-L1 overexpressed vascular endothelial cells, and the functional status of vascular endothelial cells was detected by lumen formation, Transwell cell migration assay and Ki-67 staining. Results Compared with normal skin tissues, the vascular structure and function in keloid tissues were disordered and abnormal, and the expression of PD-L1 in vascular endothelial cells was significantly decreased with regional differences. After co-culture with T cells, the lumen formation of vascular endothelial cells in the overexpressed PD-L1 group increased, the migration efficiency increased, and the proliferation was vigorous. Conclusion The expression of PD-L1 in vascular endothelial cells of keloid tissues decreased with regional differences. With the participation of T cells, the high expression of PD-L1 in vascular endothelial cells can promote the proliferation, migration and lumen formation of vascular endothelial cells, which may increase the risk of keloid formation.
