Study on the temporal relationship between the pain-related anxiety and depression and the activation of neurons in medial prefrontal cortex and anterior cingulate cortex in mice

Abstract
Objective To investigate the temporal variations of neuronal activation in emotion-related brain regions such as medial prefrontal cortex ( mPFC) and anterior cingulate cortex (ACC) during neuropathic pain accompanied by negative emotional changes in mice. Methods C57BL / 6 male mice were randomly divided into control group (CTR group) and spared nerve injury (SNI) group. Behavioral tests and immunofluorescence staining were performed on the two groups of mice at different time points after SNI intervention. Especially in mPFC and ACC brain regions, the neuron activation marker c-Fos was stained and quantified, and the types of activated neurons were further analyzed. Results Compared with the CTR group, mice in the SNI group showed anxiety-like behaviors at 28 d and depression-like behaviors at 56 d after SNI modeling. Meanwhile, the degree of activation in mPFC and ACC brain regions also showed the characteristics of temporal variations. After 3 d of SNI modeling, neurons in mPFC and ACC brain regions showed significant activation compared with those of CTR group, which was indicated by the significantly increased number of c-Fos positive neurons. Then the degree of activation gradually decreased with the progression of pain, as showed by the gradually decreased number of c-Fos positive neurons. At 56 d after SNI modeling, there was no significant difference in the number of c-Fos positive neurons in mPFC and ACC brain regions between the two groups. Correlation analysis showed that the occurrence of anxiety-like emotions was negatively correlated with the degree of mPFC neuronal activation. Further experiments showed that mPFC and ACC brain regions activated both excitatory and inhibitory neurons in the early pain stage at 3 d after SNI modeling. Conclusion The activation of neurons in mPFC and ACC brain regions changes with the progression of pain. In particular, the activation of neurons in mPFC brain region is negatively correlated with the occurrence of anxiety emotions after pain, which may play a protective role in the change and progression of pain-related negative emotions.
