The possible paracrine effect of vascular endothelial growth factor C on permeability of glomerular filtration barier in children with nephrotic syndrome

Journal Title: Przegląd Pediatryczny - Year 2006, Vol 36, Issue 3

Abstract

Introduction: The histological background of nephrotic syndrome is based on alterations of filtration membranę. Its uniąue struc-ture is dependent on visceral epithelial cells of Bowman's capsule (podocytes), basement membranę and fenestrated endothelium. Every element listed above may be impaired in the course of inflammation process which involves secretion of numerous pro-in-flammatory cytokines. The crucial role in this pathomechanism is played by different interleukins and vascular endothelial growth factor (VEGF). The aim of the study: The aim of the present research was immunohistochemical evaluation of VEGF-C expres-sion in renal glomeruli in children with nephrotic syndrome and the attempt of estimation of its correlation with clinical course of the disease. Materiał and methods: Materiał consisted of renal biopsies obtained according to WHO guidelines from children ho-spitalized in the Department of Pediatrie Cardiology and Nephrology, Poznań University of Medical Sciences in Poland. VEGF-C expression was evaluated with the use of the immunohistochemical procedurę (avidin-biotin complex) and mouse anti-human anti-VEGF-C antibodies. One-way ANOVA and Tukuey tests were employed in statistical analysis. Results: VEGF-C expression was present exclusively in podocytes. The progression of histological changes in renal glomeruli of children with nephrotic syn-drome was associated with significantly increased expression of VEGF-C in podocytes (p
